They make memory cells when they encounter a new antigen that they can defeat. For instance, if you get sick and it's a new strain of virus, then after you get better there will be memory cells that last for 7-15 years, so if you encounter that strain again those cells immediately recognize it and then signal for the reproduction of more cells...I hope that helps...They have protein structures that recognize a specific antigen on the outside of the cell.
